Hanover and Stuttgart are to be served by the Iberia group from 26 October.
Iberia Express, the group's Spanish low-cost arm, will operate to both of the German destinations from Madrid.
The frequency will be thrice weekly to Hanover and four times weekly to Stuttgart.
In October 2013, the Iberia group resumed its Madrid-Berlin service, and it currently operate 268 flights a week between Spain and the German cities of Berlin, Dusseldorf, Frankfurt and Munich.
The additional German routes are part of a batch of newly launched services that spans Amsterdam, Athens, Istanbul, Montevideo, Santo Domingo and Stockholm.
